An axon initial segment sodium channel Nav1.2 regulates sleep homeostasis

Abstract

The slow-wave activity of non-rapid-eye-movement sleep reflects system-wide oscillation of cortical neurons and sleep need, which underlies a tight genetic control. Here, we identify a mutation in sodium channel Nav1.2 that affects channel’s slow inactivation and daily sleep need in mice. Moreover, this mutation leads to markedly reduced phosphorylation changes of brain proteome and rebound of slow-wave activity after sleep deprivation, highlighting a role of Nav1.2 in sleep homeostasis regulation.
